I would invite you to spend a little time exploring the menus, as well as using the forums search before creating a new thread for each question you have.
Shipping depends on the type of shipping module you are using. The default ones are:
Flat - one price no matter what is in the cart
Per item - a set amount multiplied by the number of items in the cart
Zone - Weight-based on different geographical destinations. (i.e. states, countries, etc)
Search for "shipping" or "Zone shipping" in the forums for more information.

You can't, not with any of the existing modules. That would have to be coded special.
Currently you can only set each item to a single rate, using per item or base the shipping price on total cart weight.
Per Item example:
set per item rate to $2.00
add 10 items to your cart
each item gets a $2.00 charge for a total of 10x$2.00 = $20.00
